PolyAn 3D-aldehyde glass slides are manufactured with high substrate flatness and surface smoothness to ensure a uniform hybridization layer. They are usually used for covalent coupling of Amine-containing (bio)molecules.
The glass has low background fluorescence to enhance signal detection and improve data quality. They are suitable for both low-density and high-density microarray applications, including those used in high-throughput screening.
These slides are compatible with all microarray printers and slide scanners / imagers. We recommend the axiREADERs for both colorimetric and fluorometric applications.
Surface Modification
PolyAn’s 3D-functionalised microarray slides are functionalized with a three-dimensional (3D)-surface chemistry comprised of a long-chain polymer containing a defined number of reactive groups. This polymer is covalently linked to the surface of the slide.
PolyAn 3D-aldehyde glass slides' reactive groups consists of aldehyde groups, designed to form covalent bonds with primary amines found on most biomolecules.
Aldehyde groups react with amines, hydrazines, and aminoalkoxyacetyl-modified biomolecules, initially forming unstable imines (Schiff bases). These imines can be stabilized by reduction with NaBH4 or NaCNBH3 to yield stable amines. After coupling, unreacted aldehydes should be blocked using small molecules that penetrate the 3D matrix and quench remaining reactive sites.
Aldehyde slides are often used for amino-linked DNA probe immobilization in applications like gene expression analysis, genotyping, and other genomic studies.
Technical specifications
Dimensions: The slides are produced with standard dimensions of 25.0 x 75.6 x 1 mm.
Packaging: The slides are packaged in inert atmospheric boxes, with a capacity of 25 or 5 slides per box.
Storage: PolyAn’s aldehyde slides are characterized by a long shelf-life when stored dry, at 4-8°C and protected from sunlight. All slides are packaged in boxes under Argon atmosphere to avoid contamination with particles. The Argon atmosphere also minimizes degradation of the reactive surface through contact with air or humidity.
